Fms-like tyrosine kinase receptor 3 (FLT3), also known as CD135, is a member of the class III receptor tyrosine kinase (RTK) family. It plays a critical role in hematopoiesis by regulating the proliferation, differentiation, and survival of hematopoietic progenitor cells. FLT3 is primarily expressed in early myeloid and lymphoid progenitors and to a lesser extent in more mature monocytic lineages. Dysregulation or mutation of FLT3 is strongly implicated in acute myeloid leukemia (AML). Two main types of activating mutations are observed: Internal Tandem Duplication (ITD) and Tyrosine Kinase Domain Mutation (TKD), both resulting in constitutive activation and uncontrolled proliferation/survival signals leading to leukemogenesis.
