FMS Proto-oncogene Tyrosine-Protein Kinase (CSF1R) is a receptor tyrosine kinase that plays a crucial role in regulating the survival, proliferation, and differentiation of myeloid lineage cells, particularly macrophages. It is activated by its ligands, CSF-1 and IL-34, leading to downstream signaling through PI3K/Akt, MAPK/ERK, JNK, and SRC pathways. CSF1R is implicated in various diseases, including ALSP leukoencephalopathy and myeloid malignancies, and is a therapeutic target for cancer, neurodegenerative diseases, and inflammatory bone diseases.
