Fms-related tyrosine kinase 3 receptor (FLT3) is a class III receptor tyrosine kinase that plays a critical role in hematopoiesis. It is expressed almost exclusively in hematopoietic tissues, particularly on early hematopoietic progenitor cells. Activation by its ligand, FLT3LG, promotes survival, proliferation, and differentiation of these cells. Activating mutations in FLT3 are frequently found in acute myeloid leukemia (AML) and are associated with poor prognosis. Several small-molecule inhibitors targeting mutant or overactive FLT3 have been developed for cancer therapy, but resistance mechanisms can limit their long-term efficacy.
