Folate receptor beta (FRβ) is a high-affinity folate-binding protein encoded by the FOLR2 gene. It mediates the transport of folates into cells through endocytosis. FRβ is upregulated on immunosuppressive tumor-associated macrophages (TAMs) and myeloid-derived suppressor cells (MDSCs), making it a promising target for cancer immunotherapy.
Drugs targeting FRβ can selectively reprogram TAMs/MDSCs from tumor-promoting M2-like phenotypes toward pro-inflammatory M1-like states. Antifolates exploit this pathway for selective drug delivery.
