Overview

Follicle-stimulating hormone subunit beta (FSHB) is the hormone-specific beta subunit of follicle-stimulating hormone (FSH), a heterodimeric glycoprotein central to vertebrate reproductive regulation[3][7]. FSH is composed of a common alpha subunit and a unique beta subunit (FSHB), both necessary for hormone activity, but the beta subunit provides biological specificity and receptor binding[3][4][6][7]. The FSHB gene (chromosome 11p13 in humans) is expressed in pituitary gonadotrope cells and regulated by factors including GnRH, inhibin, and activin[4][6][7]. FSH, through its beta subunit, binds and activates the FSH receptor (a G protein-coupled receptor) on ovarian granulosa cells and testicular Sertoli cells[5][8], promoting ovarian follicle maturation and spermatogenesis, respectively[3][4][6][7]. Genetic variants in FSHB can influence risk of infertility, altered puberty onset, and reproductive pathologies[7]. Recombinant forms of FSH, containing FSHB, are clinically used for controlled ovarian stimulation and treatment of infertility; antagonists and antibodies targeting FSHB or its pathway are in experimental stages for treating metabolic and bone disorders[4][6]. FSHB levels can be measured as a biomarker for reproductive axis disorders, with therapeutic use carrying risks for ovarian hyperstimulation and multiple pregnancies[4].
