Overview

The foot-and-mouth disease virus capsid protein VP3 is a structural component of the viral icosahedral capsid that plays dual roles in viral assembly and immune evasion. As part of the pseudo-T=3 symmetric capsid structure alongside VP0 and VP1, VP3 contributes to the overall architecture and stability of the virion. Beyond its structural function, VP3 acts as a potent antagonist of host innate immunity by suppressing interferon signaling pathways through interactions with key host proteins including JAK1 and VISA, thereby facilitating viral replication and persistence. The protein is relatively conserved across FMDV serotypes, with specific residues such as arginine 56 being critical for both virulence and receptor binding. While VP3 is not currently a therapeutic drug target, understanding its mechanisms of immune evasion and structural functions remains important for developing strategies to control foot-and-mouth disease in livestock populations.
