Overview

Forkhead-associated domain-containing protein 1 (FHAD1) is a nuclear protein encoded by the FHAD1 gene, primarily distinguished by the presence of a forkhead-associated (FHA) domain, coiled-coil regions, and additional functional motifs[1][2]. The protein is predicted to be involved in transcriptional regulation and DNA repair through its phosphopeptide-binding FHA domain, which is capable of recognizing phosphorylated serine, threonine, or tyrosine residues[1][2][3][5]. In mammals, FHAD1 is highly expressed in the testis and is linked to the regulation of meiotic cell division and sperm motility, as demonstrated in knockout mouse studies where its absence led to reduced sperm motility and increased apoptosis during early spermatogenesis, but did not result in infertility[2]. FHAD1 may play a role in DNA-damage response cascades via phosphorylation-dependent protein interactions common to proteins with FHA domains[2][3][5]. No direct evidence currently supports its therapeutic targeting or druggability, and the protein has not been identified as the molecular target of any approved drugs or biomarker assays[2][6].
