Overview

Forkhead box C1 (FOXC1) mRNA is the transcript encoding the FOXC1 transcription factor, a member of the forkhead box family characterized by a distinct DNA-binding domain (1.1.1, 1.2.1). It plays a pivotal role in embryonic development, particularly in the formation of the eye's anterior segment, heart, and kidneys (1.2.1, 1.2.3). In oncology, FOXC1 mRNA is frequently overexpressed in aggressive cancers such as basal-like breast cancer (BLBC) and non-small cell lung cancer (NSCLC), where it promotes the epithelial-mesenchymal transition (EMT), cell migration, and metastasis (1.2.2, 1.3.1, 1.4.1). As a therapeutic target, FOXC1 mRNA is being investigated using RNA interference (RNAi) strategies, including small interfering RNAs (siRNAs) and short hairpin RNAs (shRNAs), to silence its expression and inhibit tumor progression (1.3.4, 2.1.1). While no clinical drugs are currently approved, its role as a prognostic biomarker and its involvement in drug resistance make it a high-priority target for nucleic acid-based therapeutics (1.4.2, 2.1.3). Challenges in targeting FOXC1 mRNA include ensuring tissue-specific delivery and avoiding developmental toxicity due to its essential role in normal organogenesis (1.3.2, 1.4.2).
