Overview

Forkhead box D3 antisense RNA 1 (FOXD3-AS1) is a long non-coding RNA transcribed antisense to the FOXD3 gene, located at chromosome 1p31.3. FOXD3-AS1 acts as a competitive endogenous RNA and regulates expression of key genes involved in cell proliferation, apoptosis inhibition, migration, and chemoresistance, frequently by sponging tumor-suppressive microRNAs and modulating epigenetic marks such as H3K27 acetylation at gene promoters. Aberrant upregulation of FOXD3-AS1 is associated with malignancy and poor prognosis in a variety of cancers—including lung, nasopharyngeal, breast, colon, and thyroid carcinomas—and serves as a promising biomarker for cancer diagnosis and prognosis. Experimental FOXD3-AS1 knockdown represses cancer cell growth, invasion, and resistance to chemotherapy, although no approved drugs currently target FOXD3-AS1 directly[1][2][3][4][5].

Mechanism of action

Experimental inhibition of FOXD3-AS1 (e.g., via siRNA, shRNA, or antisense oligonucleotides) suppresses proliferation, migration, invasion, and chemoresistance in cancer models. It also epigenetically modulates gene expression by recruiting H3K27ac to promoters.
