FoxP3 is a transcription factor that is critical for the development, stability, and function of regulatory T cells (Tregs). Tregs play a central role in maintaining immune homeostasis, self-tolerance, and preventing autoimmune responses. FoxP3 acts as a lineage specification factor for Tregs, governing their genetic program and mediating their suppressive function. Deficiency or dysfunction in FoxP3 leads to severe systemic autoimmunity. Therapeutic manipulation aims at either enhancing FoxP3/Treg function (to treat autoimmunity) or inhibiting it (to boost anti-cancer immunity).
