Overview

Forkhead box protein A1 is a DNA-binding transcription factor characterized by a forkhead domain that enables binding to the major groove of DNA and direct contact via its "wing" structures. As a "pioneer factor," FOXA1 facilitates the opening of compacted chromatin, allowing other transcription regulatory proteins, such as hormone receptors, to access DNA. It plays crucial roles in the differentiation and function of endodermal organs (liver, pancreas, lung, prostate) and is essential during development. FOXA1 is a pivotal regulator of hormone receptor signaling in breast and prostate tissue, heavily studied for its involvement in tumor progression, response to therapy, and cancer resistance mechanisms. FOXA1 is frequently mutated in prostate cancer, and its expression correlates both with favorable response in some breast tumors and with resistance and poor prognosis in others
