Overview

Forkhead box protein B2 (FOXB2) is a member of the forkhead box (FOX) family of transcription factors, defined by its role in sequence-specific DNA binding and regulation of transcription by RNA polymerase II. FOXB2 is implicated in both development and oncogenesis, with context-dependent functions. In certain cancer contexts, such as advanced prostate cancer, FOXB2 acts as a potent activator of Wnt signaling by inducing expression of several Wnt ligands, notably WNT7B, driving neuroendocrine differentiation and malignant progression. This mechanism does not rely on classical Wnt co-receptors and involves key transcriptional cofactors such as YY1, JUN, and DDX5. Conversely, in pancreatic ductal adenocarcinoma, FOXB2 is epigenetically silenced, and its expression has tumor-suppressive effects, inhibiting β-catenin and reducing stem-like malignancy. FOXB2 is also active during embryogenesis, with distinct expression dynamics suggesting a role in neural patterning and organogenesis[1][4].
