Overview

Forkhead box protein D1 (FOXD1) is a member of the forkhead box (FOX) family of transcription factors defined by a distinct DNA-binding forkhead domain. FOXD1 is essential for organogenesis, especially the development and morphogenesis of the kidney, retina, and optic chiasm. It plays critical roles in nephron progenitor differentiation, regional identity assignment in neural tissues, and regulation of cellular reprogramming. In cancer biology, FOXD1 is implicated as an oncogene, where increased expression correlates with higher tumor grade, poor prognosis, and chemoresistance. FOXD1 also helps regulate inflammatory reactions and prevent autoimmunity. Its broad developmental and cellular repertoire makes FOXD1 both a key biomarker and a putative therapeutic target in oncology and regenerative medicine, but also raises concerns for safe targeting given its essential role in normal tissue patterning and cell fate specification.
