Overview

Forkhead box protein D3 (FOXD3) is a DNA-binding transcription factor of the forkhead box (FOX) family, characterized by the forkhead domain[1][5]. FOXD3 serves as a pivotal regulator of early development, pluripotency, neural crest lineage specification, and acts as a tumor suppressor. It functions as both a transcriptional repressor and activator, regulating gene expression and priming enhancers during development by recruiting chromatin remodelers and histone deacetylases[2][3]. In embryogenesis, FOXD3 maintains stem cell pluripotency and prevents premature differentiation. In the neural crest, it determines lineage fate and maintains progenitor pools. In pathology, FOXD3 loss or epigenetic silencing is associated with several diseases, including autoimmune disorders and cancers such as melanoma and gastric carcinoma[2][3].
