Overview

Forkhead box protein J2 (FOXJ2) is a transcriptional activator belonging to the forkhead box family of transcription factors, characterized by a conserved forkhead (winged-helix) DNA binding domain[1][2][3][6]. It binds to specific DNA sequences and regulates the expression of multiple genes, functioning as a regulator of biological processes such as angiogenesis and cell differentiation[1][6]. FOXJ2 plays an important role in tumorigenesis, progression, and metastasis of certain cancers—implicated, for example, in inhibition of breast cancer metastasis through regulation of EMT markers E-cadherin and vimentin[2][4]. In mice, FOXJ2 is required for the production of interferon-alpha by plasmacytoid dendritic cells, highlighting a potential role in immune regulation[5]. There are currently no widely reported small-molecule or biological drugs directly interacting with or targeting FOXJ2, nor are mechanisms of action, biomarkers, or safety concerns established for clinical targeting of FOXJ2.
