Overview

Forkhead box protein J3 (FOXJ3) is a member of the forkhead box (FOX) family of transcription factors, characterized by a conserved forkhead DNA-binding domain[1][2]. It regulates gene expression by binding to specific DNA sequences as a transcriptional activator and is primarily active in the nucleus. FOXJ3 is implicated in the regulation of cell proliferation, differentiation, and tissue development. In the nervous system, it is important in oligodendrocyte precursor cells, linking exit from cell cycle to differentiation. In skeletal muscle, FOXJ3 is pivotal for myofiber identity and efficient muscle regeneration, partly by activating MEF2C and regulating mitochondrial biogenesis[1][2]. It is also essential for spermatogenesis, the survival of spermatogonia, and progression of meiosis in spermatocytes[2][3][6]. In cancer, FOXJ3 appears to function as a tumor suppressor whose downregulation can promote proliferation and invasion of cancer cells. The expression and function of FOXJ3 are subject to regulation by microRNAs[1]. No established drugs, mechanisms of action, or biomarker roles are documented for FOXJ3, and it is not considered a direct therapeutic target at present.
