Forkhead box protein L3 (FOXL3) is a DNA-binding transcription factor belonging to the forkhead box family, predicted to regulate transcription of specific genes by binding cis-regulatory DNA regions, particularly by RNA polymerase II[3][7]. It is a key determinant of sexual fate in the germline of certain vertebrates, notably teleost fish, where it orchestrates commitment of germ cells to spermatogenesis or oogenesis by suppressing spermatogenesis in females and participating in sex-specific meiotic and folliculogenic pathways[1][6]. FOXL3 expression exhibits sexual dimorphism in the gonads of fish, and its ablation can result in altered germ cell fate, such as the appearance of functional sperm in female gonads[1][6]. In humans, FOXL3 is a protein-coding gene with limited characterized function, and is not a currently established therapeutic or drug target[3][7]. Its disease association is limited, with reported links to hermaphroditism and orofacial cleft 1[3].
