Overview

Forkhead box protein N3 (FOXN3) is a member of the forkhead/winged-helix family of transcription factors encoded by the FOXN3 gene on human chromosome 14q31.3-q32.11[1][5]. FOXN3 primarily acts as a transcriptional repressor and has critical roles in cell cycle control, particularly in the DNA damage response where it mediates cell cycle arrest and facilitates DNA repair to maintain genomic stability[2][3][5]. FOXN3 can suppress tumor proliferation through blocking cyclin-dependent kinases and modulating the AKT/MDM2/p53 signaling pathway[4][5]. It is frequently downregulated in various cancers, where reduced FOXN3 expression correlates with poorer prognosis[2][4][5]. However, FOXN3 also has context-dependent roles and can act as an oncogene in specific cancers such as pancreatic cancer[2]. It forms complexes with cofactors, such as NEAT1 and SIN3A, affecting gene expression networks involved in cancer cell migration and metastasis[2]. No drugs directly targeting FOXN3 are currently known, and it is not recognized as a routine pharmacological target; however, FOXN3 expression is being investigated as a prognostic biomarker in select malignancies[2][3][4][5].
