Forkhead box protein O4 (FOXO4)

Forkhead box protein O4 (FOXO4) is a transcription factor that belongs to the forkhead family. It regulates gene expression by binding to specific DNA sequences and is modulated by post-translational modifications. FOXO4 plays key roles in oxidative stress response, longevity pathways, insulin signaling, cell cycle progression, apoptosis, and DNA repair. It is involved in various biological processes, including insulin receptor signaling, DNA damage checkpoint signaling, and muscle organ development. Alterations in FOXO4 are associated with cancer and age-related diseases, making it a potential therapeutic target.

Modulation of FOXO4 activity via post-translational modifications (phosphorylation, ubiquitination, acetylation) to regulate gene expression; disruption of FOXO4 interaction with p53 to induce apoptosis in senescent cells.
