Target intelligence / Profile preview
Forkhead box protein O6 (FOXO6) is a DNA-binding transcription factor belonging to the FOXO subfamily, characterized by a conserved forkhead (winged helix) domain[4][5][7]. FOXO6 is expressed predominantly in the brain (hippocampus, cingulate cortex, nucleus accumbens, amygdala), liver, and skeletal muscle[2][3][6][8], regulating processes such as gluconeogenesis during fasting, neuronal differentiation/memory, and cardiac remodeling under pathological conditions[1][3][6]. FOXO6 activation or nuclear localization is typically controlled by phosphorylation via the AKT/PKB pathway—regulation is more restricted compared to other FOXO family members due to its unique structure and tissue distribution[2][4]. In pathophysiological states, FOXO6 is upregulated and promotes heart failure by facilitating cardiac fibrosis and hypertrophy via activation of genes like KIF15 and TGF-β1[1], and in insulin resistance, FOXO6 drives excess gluconeogenesis, contributing to adverse metabolic outcomes[3][5].
Drugs or molecules affecting FOXO6 act by modulating its nuclear localization, phosphorylation state (AKT/PKB pathway), or expression, altering gene transcription for processes like gluconeogenesis or fibrosis
