Overview

Forkhead box protein P4 (FOXP4) is a nuclear transcription factor encoded by the FOXP4 gene, belonging to the forkhead box family, subfamily P. It possesses a winged helix/forkhead DNA-binding domain and regulates the transcription of genes required for cell proliferation, differentiation, and survival during both embryonic development and adult tissue maintenance. FOXP4 plays key roles in development of the nervous, pulmonary, immune, and reproductive systems. It is a tissue- and cell type-specific regulator whose dysfunction is implicated in male infertility (through defective spermatogonial stem cell renewal), oncogenesis in multiple cancer types, and neurodevelopmental disorders. FOXP4 is regulated both at the gene and post-transcriptional level, notably by noncoding RNA networks, and modulates downstream signaling pathways such as Wnt and PI3K/AKT/mTOR. FOXP4 is under study as both a disease biomarker and a potential, though challenging, therapeutic target.

Mechanism of action

Not applicable (no direct drugs reported). Mechanistically, small RNAs (such as microRNAs) and long noncoding RNAs regulate FOXP4 expression, affecting oncogenic and developmental pathways including Wnt and PI3K/AKT/mTOR signaling. MiRNAs that target FOXP4 have tumor suppressor activities through inhibition of FOXP4 function.
