Overview

Forkhead box protein R2 (FOXR2) is a member of the forkhead box (FOX) family of transcription factors, characterized by a conserved forkhead DNA-binding domain and mainly localized in the nucleoplasm[1][2][5][7]. In humans, FOXR2 is encoded by a single gene on the X chromosome (Xp11.21) with expression physiologically restricted to the testis, but it is aberrantly overexpressed in a wide range of malignancies, where it acts as an oncogene. FOXR2’s oncogenic functions include enhancement of cell proliferation, inhibition of apoptosis, increased metastatic potential, and modulation of multiple oncogenic signaling pathways, including MYC, Wnt/β-catenin, and ETS transcription circuitries. Its pivotal role in many cancers makes FOXR2 both a potential prognostic biomarker and an emerging but largely unexplored therapeutic target[1][6].
