Overview

Forkhead box protein S1 (FOXS1) is a DNA-binding transcription factor of the forkhead box (FOX) family characterized by a winged helix domain, encoded by an intronless gene and expressed most prominently in the aorta and kidney. FOXS1 is a context-dependent regulator of gene expression, acting as a transcriptional repressor and modulator of cellular proliferation, migration, epithelial–mesenchymal transition, and differentiation; its role varies by tissue and cancer type, functioning as both an oncogene and a tumor suppressor in different settings. FOXS1 participates in key signaling pathways, influences the tumor immune environment, and is implicated in carcinogenesis, fibrosis, and vascular organization. There are no approved drugs directly targeting FOXS1, but altered FOXS1 expression is associated with several disease states and may serve as a prognostic marker for cancer progression and immune landscape.
