The formation of a protective barrier on the oral mucosa is a multifaceted biological process consisting of the physical (epithelial cell layers with tight junctions), microbiological (commensal microbiota), and immunological (mucosal immune system) defenses of the oral cavity[1][2][3]. The oral epithelial barrier is composed of stratified squamous epithelial cells, cell–cell junctions (tight junctions, desmosomes, adherens junctions), basement membrane, and the underlying lamina propria[1][3]. This barrier is constantly renewed and interacts dynamically with saliva-derived mucus, commensal and pathogenic microorganisms, and immune cells to prevent microbial invasion, physical injury, and chemical insult, while allowing selective transport and immune surveillance[1][2][3]. Disruption of this barrier is implicated in various diseases such as oral infections, inflammation, and cancer[1]. The formation of a protective barrier is therefore an emergent tissue property, not a druggable or canonical "target" in the sense of a receptor, enzyme, or molecular pathway component[1][2][3].
