Overview

Formin-1 (FMN1) is a member of the formin family of proteins, which are highly conserved actin nucleating and organizing factors essential for defining cell shape, polarity, migration, and division[1][3][4][6]. Formin-1 is characterized by formin homology domains (FH1 and FH2) that mediate its ability to nucleate and elongate unbranched actin filaments. The FH2 domain specifically encircles the barbed (fast-growing) end of actin filaments, promoting polymerization while protecting filaments from capping proteins[4][5][6]. FMN1 also acts as a scaffold, interacting with proteins like profilin and α-catenin to coordinate cytoskeletal and cell adhesion networks[3]. In mouse, Fmn1 is essential for limb and kidney morphogenesis, and mutations lead to the \"limb deformity\" phenotype. FMN1 is encoded on human chromosome 15q13.3[3]. Although not a classical drug target or receptor, FMN1 is a critical mediator of cellular architecture and motility, with developmental roles supported by both animal and human data[1][3]. There are currently no known drugs directly targeting FMN1, and its therapeutic tractability remains largely unexplored.
