Formins are important regulators of the actin and microtubule cytoskeleton. The FH2 domain, in particular, binds actin filament barbed ends and catalyzes nucleation and elongation of unbranched actin filaments[1][4][5][7]. Other domains (FH1, FH3, etc.) are involved in profilin binding, cellular localization, or regulatory functions. The term "formin domain" does not refer to a specific protein, enzyme, receptor, or therapeutic target, but rather to a conserved sequence found in a large family of proteins (formins). While formin proteins (such as DIAPH1, mDia1, or Bni1) can themselves be valid research or drug targets, the domain alone is not targetable and is generally not used in therapeutic contexts.
