Overview

Formyl peptide receptor 1 (FPR1) is a high-affinity G protein-coupled receptor (GPCR) primarily expressed on the surface of phagocytic cells, such as neutrophils and macrophages (UniProt: P21462). It serves as a critical pattern recognition receptor in the innate immune system, specifically detecting N-formylated peptides derived from bacteria or damaged mitochondria (PubMed: 29334381). Upon binding its prototypical ligand, fMLP, FPR1 triggers intracellular signaling that leads to chemotaxis, phagocytosis, and the release of reactive oxygen species (IUPHAR/BPS Guide to Pharmacology). While essential for host defense, dysregulated FPR1 activity is implicated in chronic inflammatory conditions, ischemia-reperfusion injury, and certain malignancies (PubMed: 30635471). Consequently, FPR1 is a significant therapeutic target; antagonists are being developed to mitigate excessive inflammation, while agonists are explored for promoting wound healing and resolving infections (StatPearls: StatPearls Publishing).
