Overview

The Fos–Jun–DNA complex, commonly called the AP-1–DNA complex, comprises a heterodimer of the bZIP transcription factors Fos and Jun bound to a specific DNA sequence known as the AP-1 binding site[2][3][4][5]. The complex regulates transcription of genes involved in diverse processes such as proliferation, differentiation, and apoptosis[4]. Structurally, Fos and Jun dimerize via leucine zipper motifs, juxtaposing their basic regions for sequence-specific DNA binding[5]. The activity and DNA-binding ability of the complex can be modulated by redox state and interaction with other transcription factors, as well as post-translational modifications[3][5]. Aberrant regulation or overactivation of AP-1 (Fos/Jun) is implicated in cancer, inflammation, and other diseases, making AP-1 a target of research for new drugs, though no direct therapies for the complex are approved[4]. The Fos–Jun–DNA complex itself is not considered a classical therapeutic target, but the AP-1 transcription factor is under investigation as a modulator of pathophysiological gene expression.
