Fos-related antigen 2 (FOSL2) is a leucine zipper-containing transcription factor that forms part of the dimeric AP-1 complex after heterodimerization with JUN proteins. It regulates gene expression crucial for cell proliferation, differentiation, immune specification (notably restraining Th17 cell fate), and transformation. FOSL2 is implicated in numerous physiological and pathological processes, including cancer progression, autoimmune disorders, cardiac development, fibrosis, and osteoclast function. Its activity is tightly regulated via context-dependent dimerization and DNA affinity, which is affected by disease-associated single nucleotide polymorphisms (SNPs). While not the direct target of approved drugs, FOSL2 remains a significant node for disease research and potential therapeutic intervention.
