Overview

Four and a half LIM domains protein 2 (FHL2) is a multifunctional, LIM-domain-only scaffolding protein encoded by the FHL2 gene, usually referred to as FHL2[1][2][3]. FHL2 contains four complete and one half LIM zinc finger domains, enabling extensive protein-protein interactions and the assembly of multi-protein complexes. It primarily localizes to focal adhesions, the actin cytoskeleton, and the nucleus, where it acts as a mechanotransducer—sensing mechanical signals from the extracellular matrix and regulating downstream gene expression in a tension-dependent manner[2]. In the nucleus, FHL2 functions as a co-factor with various transcription factors (such as AP-1, NF-κB, androgen receptor) to control genes implicated in cell proliferation, adhesion, and survival[2][3]. FHL2 has been implicated in diverse roles in cancer, functioning either as a tumor suppressor or an oncogene depending on context, and its expression is dynamically regulated in response to mechanical and biochemical cues[3][1]. Its broad tissue distribution and involvement in key intracellular signaling make it a biologically significant but therapeutically complex target.
