FOXD2 adjacent opposite strand RNA 1 (FOXD2-AS1) is a long non-coding RNA with no known protein product, widely upregulated in various cancer types and implicated in tumor progression, chemoresistance, cancer stemness, and recurrence. It primarily functions as a competing endogenous RNA (ceRNA), acting as a molecular sponge for tumor-suppressor microRNAs including miR-206 and miR-7-5p, resulting in increased expression of oncogenic proteins such as annexin A2 (ANXA2) and telomerase reverse transcriptase (TERT)[1][2][3][4][5]. FOXD2-AS1 also scaffolds protein complexes (e.g., STAT3 and PRMT5) to promote oncogenic transcriptional programs and can repress tumor suppressors like TRIB3 to activate proliferative signaling. High FOXD2-AS1 expression correlates with poor prognosis and contributes to resistance to chemotherapy in multiple tumor models. At present, there are no approved drugs targeting FOXD2-AS1, but its central role in regulating oncogenic networks makes it a candidate biomarker and potential future therapeutic target in oncology.
