FOXP1 intronic transcript 1 (FOXP1-IT1) is a long non-coding RNA located within an intron of the FOXP1 gene, specifically on human chromosome 3 (GRCh38.p12). It exists in both spliced (3.4 kb) and unspliced (4.0 kb) forms. FOXP1-IT1 directly binds to histone deacetylase 4 (HDAC4) and modulates its phosphorylation state, thereby influencing the transcriptional activity of FOXP1—particularly via the P1 promoter. This lncRNA counteracts integrin Mac-1 clustering-induced downregulation of FOXP1, attenuates IRAK1-mediated repression, and suppresses monocytic differentiation and cell adhesion in vitro. FOXP1-IT1 represents an important epigenetic regulatory mechanism in immune cell function and offers potential new targets for anti-inflammatory therapy development, though its status as a therapeutic target is not established.
