Friend leukemia integration 1 transcription factor (FLI1) is a proto-oncogene and a member of the ETS family of transcription factors encoded by the FLI1 gene. It regulates transcription of many genes involved in cell proliferation, differentiation, and survival, with a primary role in hematopoietic and endothelial cell development[1][3][4]. FLI1 is critical for maintaining and differentiating hematopoietic stem cells, controlling vasculogenesis and angiogenesis, and regulating blood cell and platelet formation. Aberrant expression or chromosomal translocation involving FLI1, especially the EWS-FLI1 fusion, is a major driver of Ewing sarcoma and is implicated in leukemias, autoimmune diseases, and platelet dysfunction disorders. FLI1 is considered a promising therapeutic target in oncology and autoimmunity, though challenges remain due to the difficulty in developing specific transcription factor inhibitors[3][1][4][2][5][6].
