Overview

Frizzled-2 receptor (FZD2) is a member of the Frizzled gene family and encodes a 7-transmembrane G protein-coupled receptor that primarily serves as a receptor for Wnt signaling proteins[7]. FZD2 is involved in both canonical (β-catenin-dependent) and non-canonical (Wnt/PCP and Wnt/Ca^2+^) signaling pathways, regulating diverse cellular processes including embryonic development, cellular proliferation, epithelial morphogenesis, and cell fate decisions[2][4][6]. The receptor is developmentally regulated, with high expression in fetal tissues such as kidney and lung, and in adult colon and ovary[7]. Mutations in FZD2 are associated with developmental disorders (e.g., omodysplasia), and FZD2 has been implicated in the growth and vascularization of certain tumors, notably neuroblastoma, suggesting its emerging importance as a therapeutic target[2][4][8]. Experimental blockade of FZD2 in cancer models inhibits tumor growth and disrupts key Wnt-dependent signaling cascades, although no selective clinical drugs are yet available[2].
