Frizzled-4 receptor (FZD4) is a member of the Frizzled gene family encoding a G protein-coupled receptor that serves as a core cell surface receptor in the Wnt signaling pathway[1][5][8]. It is composed of an extracellular cysteine-rich domain (CRD) for ligand (Wnt or Norrin) binding, a flexible linker, and a seven-transmembrane domain typical of GPCRs[1][7]. FZD4 is pivotal for cellular processes including proliferation, migration, adhesion, and tissue development, particularly in vascular and neural tissues[5][8]. Mutations in FZD4 are causatively linked to familial exudative vitreoretinopathy, an inherited blindness disorder. While there are no approved small-molecule drugs directly targeting FZD4, its unique ligand interactions and signal transduction mechanisms are under active investigation for therapeutic development in oncology, ophthalmology, and vascular disease[1][2][6][8].
