FZD4 encodes a cell surface receptor, a key component of the Wnt signaling pathway. This pathway regulates crucial cellular processes. FZD4 binds Norrin with high affinity, which is essential for vascular development in the retina and inner ear and specialization of retinal cells during early development. FZD4 primarily activates the canonical Wnt/β-catenin signaling pathway but can also participate in noncanonical pathways. Mutations or variants in the FZD4 gene are linked to Familial Exudative Vitreoretinopathy and dysregulated expression has been observed across different cancer types.
