Frizzled class receptor 6 (FZD6) is a member of the frizzled family of G protein-coupled receptors embedded in the cell membrane, primarily serving as a receptor for Wnt proteins[1][3][6]. FZD6 plays a crucial role in both canonical (β-catenin dependent) and non-canonical (planar cell polarity, PCP) Wnt signaling pathways, regulating vital processes such as cell proliferation, adhesion, migration, embryonic tissue patterning, and organ morphogenesis[1][2][3][4]. Its physiological significance is most evident in nail and hair development; mutations in FZD6 cause isolated autosomal recessive nail dysplasia (nonsyndromic congenital nail disorder 10)[1]. FZD6 is also implicated in various cancers, and its expression is associated with inflammatory and neoplastic conditions of the intestine and other organs[4][5]. Structurally, FZD6 has seven transmembrane domains, a N-terminal cysteine-rich domain that binds Wnt ligands, and lacks the PDZ-binding motif present in some other family members[3][2]. There are currently no approved drugs that target FZD6 directly, but the receptor is considered a promising target for modulation of Wnt signaling in several disease states[2][3].
Drugs or tool compounds targeting FZD6 would act by modulating Wnt signaling pathways, either by antagonizing or potentiating receptor-ligand interactions, thereby affecting downstream signaling such as β-catenin dependent (canonical) or planar cell polarity (non-canonical) pathways[3][2].
