Frizzled-8 (FZD8) is a seven-transmembrane domain receptor belonging to the frizzled gene family. It functions as a receptor for Wnt signaling proteins, primarily involved in the canonical Wnt/β-catenin pathway but also participating in non-canonical pathways like Planar Cell Polarity (PCP) and Wnt/Ca²⁺ signaling. Upon ligand binding, it forms complexes with co-receptors such as LRP5 or LRP6, triggering downstream transcriptional activity important for development, tissue maintenance, cell fate determination, cell polarity, tissue patterning, and proliferation. FZD8 is implicated in diseases such as renal cell carcinoma, cystitis, and Axenfeld-Rieger Syndrome type I.
