Frizzled receptors are a family of ten distinct seven-transmembrane proteins (FZD1-10) that function as the principal receptors for secreted Wnt signaling ligands (UniProt, 2023). These receptors are classified as Class F G protein-coupled receptors and are essential for coordinating complex biological processes including embryonic development, cell fate determination, and tissue homeostasis (StatPearls, 2023). Upon Wnt binding, Frizzled receptors activate either the canonical beta-catenin pathway or non-canonical pathways like the planar cell polarity (PCP) and Wnt/Ca2+ pathways (PubMed, PMID: 28273102). Aberrant activation of Frizzled signaling is a hallmark of many cancers, where it drives uncontrolled cell proliferation and stemness, and is also linked to bone density and vascular diseases (Nature Reviews Drug Discovery, 2017). Therapeutic development targeting Frizzled receptors includes monoclonal antibodies like Vantictumab and decoy receptors like Ipafricept, though clinical progress has been tempered by safety concerns regarding bone health and intestinal integrity (Journal of Clinical Oncology, 2016).
Antagonism of Wnt ligand binding to the extracellular cysteine-rich domain (CRD) of Frizzled receptors to inhibit downstream beta-catenin stabilization and TCF/LEF-mediated gene transcription (PubMed, PMID: 25135992).
