Overview

Fungal cell membrane and biofilm integrity refers to the structural and functional robustness of the fungal plasma membrane and the protective extracellular matrix produced during biofilm formation. The fungal cell membrane is a lipid bilayer primarily composed of ergosterol, which maintains fluidity and serves as a scaffold for membrane-bound proteins, distinguishing it from mammalian membranes that utilize cholesterol (Prasad et al., 2016). Biofilms are organized communities of microorganisms attached to surfaces and encased in a self-produced matrix of polysaccharides, proteins, and extracellular DNA, which acts as a physical barrier against antifungal penetration and host immune cells (Finkel & Mitchell, 2011). These structures are vital for fungal survival, environmental adaptation, and the development of antimicrobial resistance (Nobile & Johnson, 2015). Pharmacological intervention typically involves disrupting the membrane via ergosterol binding or inhibiting the enzymes responsible for ergosterol synthesis, as well as targeting biofilm-specific components to enhance drug efficacy (Odds, 2003). Maintaining the integrity of these barriers is a primary defense mechanism for pathogens like Candida and Aspergillus species during invasive infections. Disruption of these targets leads to cytoplasmic leakage, metabolic failure, and eventual cell death.

Mechanism of action

Antifungal drugs disrupt this target by binding to ergosterol to create membrane pores, inhibiting ergosterol biosynthesis enzymes (e.g., CYP51), or inhibiting the synthesis of cell wall/matrix components like 1,3-beta-glucan (Odds, 2003; Prasad et al., 2016).
