Overview

Fungal cell membrane lipids, most notably ergosterol, are essential components that maintain the structural integrity, fluidity, and permeability of the fungal cell envelope (Nature Reviews Microbiology, 2015). Ergosterol serves a functional role analogous to cholesterol in mammalian cells, facilitating the proper orientation of membrane proteins and the formation of lipid rafts (Journal of Lipid Research, 2016). Because ergosterol is absent in human cells, it represents a primary target for selective antifungal therapy (StatPearls, 2023). Polyene antibiotics, such as amphotericin B and nystatin, interact directly with these lipids to form transmembrane channels or pores (PubMed, 2014). This interaction leads to the rapid leakage of intracellular ions, such as potassium, ultimately resulting in fungal cell death (NIH, 2022). Targeting these lipids is a critical strategy for managing a wide range of fungal infections, including invasive candidiasis and aspergillosis (Clinical Microbiology Reviews, 2017).
