Fungal cell wall mannoproteins are highly glycosylated proteins that constitute the outermost layer of the fungal cell wall, serving as the primary interface between the fungus and its environment (PMID: 16460230). These proteins are essential for maintaining structural integrity and are critical for virulence, mediating adhesion to host surfaces and the formation of biofilms (PMID: 19151350). In the context of infection, mannoproteins act as pathogen-associated molecular patterns (PAMPs) recognized by host immune receptors like Dectin-2 and Mannose Receptor, though they also facilitate immune evasion by masking underlying beta-glucans (PMID: 25135932). Therapeutically, they are targeted by mannan-binding agents such as pradimicins and are the focus of monoclonal antibody and vaccine development (PMID: 17174487). Additionally, the detection of circulating mannoproteins, such as galactomannan and mannan, serves as a vital diagnostic biomarker for invasive fungal diseases like aspergillosis and candidiasis (PMID: 15561820).
Mannan-binding agents like pradimicins recognize and bind to the mannose residues of mannoproteins in a calcium-dependent manner, leading to the disruption of the fungal cell membrane and cell wall, ultimately causing cell death (PMID: 17174487). Monoclonal antibodies target specific mannoproteins to block adhesion to host tissues or to opsonize the fungal cells for phagocytosis (PMID: 22460961).
