Overview

Fungal cell wall synthesis enzymes comprise a group of essential membrane-bound and extracellular proteins responsible for the assembly and maintenance of the fungal cell wall architecture [1, 5, 7]. The primary component of this target class is (1,3)-beta-D-glucan synthase, a multi-subunit complex (encoded by FKS genes) that polymerizes glucose monomers to form the wall's structural scaffold; it is the specific target for echinocandin and triterpenoid antifungals [2, 10, 17]. Other members include chitin synthases, which generate chitin microfibrils essential for mechanical strength and septum formation, and mannosyltransferases like Gwt1, which are involved in the synthesis of the outer mannoprotein layer [11, 15, 19]. Because the fungal cell wall is absent in mammalian cells, these enzymes are ideal targets for highly selective antifungal therapy with a low potential for host toxicity [3, 8, 14, 21]. Drug-mediated inhibition compromises the wall's structural integrity, leading to osmotic fragility, cell lysis, and fungal death [12, 14].

Mechanism of action

The primary mechanism of action involves the inhibition of enzymes such as (1,3)-beta-D-glucan synthase (targeted by echinocandins and triterpenoids) or chitin synthase (targeted by nikkomycins), which prevents the polymerization of essential cell wall polysaccharides, resulting in osmotic instability and fungal cell lysis (Source: PMC, NIH, PubMed).
