Overview

The fungal cell wall and membrane are essential structural barriers that maintain cellular integrity and protect against environmental stress. The cell wall is a complex matrix primarily composed of polysaccharides such as 1,3-beta-glucan and chitin, which are synthesized by specialized enzyme complexes like 1,3-beta-glucan synthase and chitin synthase [1]. The fungal plasma membrane is uniquely characterized by the presence of ergosterol, a sterol that regulates membrane fluidity and is absent in mammalian cells [2]. Antifungal drugs exploit these differences: echinocandins inhibit glucan synthesis to cause osmotic lysis, while polyenes bind directly to ergosterol to create lethal pores [3]. Additionally, azole antifungals disrupt the ergosterol biosynthetic pathway by inhibiting lanosterol 14-alpha-demethylase [4]. Because these components are vital for fungal survival and often lack direct human homologs, they remain the primary targets for treating systemic and superficial mycoses [5].
