The fungal plasma membrane is a dynamic, selectively permeable barrier that separates the interior of the fungal cell from its external environment. It is primarily composed of a phospholipid bilayer embedded with proteins and ergosterol. The membrane plays critical roles in nutrient uptake, signal transduction, environmental sensing, and homeostasis. Key proteins include the H+-ATPase Pma1, which maintains electrochemical gradients, and glucan synthases involved in cell wall construction. The unique sterol composition, particularly ergosterol, makes it a prime target for antifungal drugs like polyenes and azoles.
Polyenes bind ergosterol, causing pore formation and membrane disruption. Azoles inhibit ergosterol biosynthesis.
