Fungal squalene epoxidase (SQLE), also known as squalene monooxygenase, is a key FAD-dependent enzyme in the ergosterol biosynthesis pathway of fungi. It catalyzes the stereospecific conversion of squalene to 2,3(S)-oxidosqualene, which is essential for producing ergosterol, the principal sterol component of fungal cell membranes. SQLE is a validated antifungal drug target, and its inhibition leads to accumulation of squalene and depletion of ergosterol, disrupting membrane integrity and function. Resistance mutations have been identified, posing a challenge for antifungal development.
