Overview

The fusion glycoprotein (F protein) is a trimeric class I viral fusion protein expressed on the surface of RSV, hMPV, and PIV3 virions. Its primary function is to mediate fusion of the viral envelope with host cell membranes during infection. The prefusion conformation is metastable and contains highly potent neutralizing epitopes, making it a superior target for therapeutic antibodies and new-generation vaccines. Engineering of disulfide bonds and cavity-filling mutations has enabled stabilization of the prefusion form, drastically improving immunogenicity and protection in preclinical and clinical studies. Several monoclonal antibodies and vaccine candidates are specifically designed to recognize and stabilize/deliver these prefusion-specific epitopes.
