FXYD domain-containing ion transport regulator 7 (FXYD7) is a member of the FXYD protein family, characterized as a single-span type I transmembrane protein expressed exclusively in the brain, in both neurons and glial cells. FXYD7 interacts with and modulates the Na⁺/K⁺-ATPase, primarily associating with the α1–β1 isozyme in brain tissue, where it decreases the apparent affinity of the enzyme complex for extracellular potassium. This fine-tuning of Na⁺/K⁺-ATPase kinetics by FXYD7 is thought to play an important role in regulating neuronal excitability and maintaining ion homeostasis. FXYD7 undergoes unique post-translational modifications, notably O-glycosylation of N-terminal threonine residues, contributing to its stability and function in the central nervous system. Clinical utility and therapeutic targeting have not yet been established, but FXYD7 is of biological interest for its specific regulatory function in brain ion transport.
