FYVE, RhoGEF and PH domain-containing protein 3 (FGD3) is a multidomain signaling protein containing FYVE (zinc finger), RhoGEF (guanine nucleotide exchange factor), and PH (pleckstrin homology) domains[1][2][3][6]. It is encoded by the human FGD3 gene and promotes the formation of filopodia—thin, actin-based protrusions involved in cell movement and sensing—by activating the small GTPase CDC42, facilitating the exchange of GDP for GTP[1][2][3][5][9]. Through its activity, FGD3 is a key regulator of actin cytoskeleton organization, cell shape, and motility, making it of therapeutic interest, particularly in diseases involving aberrant cell migration such as cancer[2][8][9]. Although no approved drugs target FGD3 directly, active drug discovery efforts focus on finding modulators that may influence its role in cytoskeletal regulation[2].
Inhibitors may act by blocking the GEF activity toward CDC42, thereby modulating actin cytoskeleton dynamics and cell morphology/motility
