FGD6 is a protein that acts as a guanine nucleotide exchange factor, activating the small GTPase Cdc42 to regulate actin cytoskeleton dynamics, coordinate cell adhesion, polarity, and membrane recycling, particularly in osteoclasts during bone degradation. It contains FYVE and two PH domains, which mediate membrane localization through phosphoinositide binding. FGD6 has been implicated in cancer biology, with its upregulation in gastric cancer correlating with poor prognosis, and in inherited syndromes such as Aarskog syndrome. While its key cellular functions and disease associations are noted, FGD6 is not currently established as a therapeutic target, nor are there drugs known to interact directly with it.
